Check them for any play. If you detect any play whatsoever they arebad and need replacing.
Dry ball joints/suspension joints. Universal joints going bad. Worn brake pad "squealers".
A good sign is when you stick it in drive or reverse and you have your foot on the brake but it still clunks when it kicks into gear

Put the car on a rack and have the ball joints inspected by a professional if you suspect they are bad. Your car will dart from side to side and handle poorly if they are bad. This is a very dangerous situation so have this looked at ASAP.
It could possibly be bad universal joints, bad transmission, damaged pinion gear in the rear end, etc. It could be caused by many possible different things.

Remove the drive shaft and take your hand and work the joints around and back and forth. If they are STIFF are hard to work are real loose with LOT"S of play then replace them. You can also remove someof the cups and make sure there not rusted and are full of grease too.
If it is a front wheel drive vehicle, it could be your cv axle or bad struts. yes bad shocks could cause vibration ,but i know my car had a bad cv axle in it a while back. the problem was the cv axle was broke in 2. Take it to an auto shop and ask them to test drive it and see if they can tell you what the problem might be. but if its a front wheel drive car, then i would bet money it is the cv axle but if its a rear wheel drive vehicle then ask them to check your universal joints(U-Joints.)
