After you collect all five sacred items, return to the Tree of Immortality. Zeus will take them. Then you can go to Hercules and talk to him. Use the Touchscreen Mirror to teleport to Hades throne room and Hercules will move the boulder.
You cannot go to Poseidon's throne room ordinarily until you have returned the 5 sacred items to the Tree of Immortality. Once you do, you can get Hercules to help you reach Poseidon and Hades. (You will need the Touchscreen Mirror from Aphrodite's test : the gods are Ares, Hermes, Aphrodite, Artemis, and Hestia.)
Hercules is not going to help you during the hunt for the Five Sacred Items. It is only after Zeus has taken them from you that he agrees. His contribution is to move the large rocks blocking you from the Hades and Poseidon throne rooms. He also smashes the lock on the Mount Olympus gate. Take his crown and teleport to the underwater place of Poseidon. Hercules will break the stone floor and you can enter Poseidon's throne room. Then teleport using the lightning sign on the mirror and Hercules will smash the lock on the gate. You'll enter Mount Olympus and Hercules will be turned to stone by Medusa. Battle Zeus at the top of Mt. Olympus and Athena will award you with the Island Medallion.
Go to Herc's Hero Hut and talk to him. After he is convinced, use the Touchscreen Mirror from Aphrodite to take you to Hades lair. Hercules will help move the boulder and you can get the Hades Crown. Then take Hercules to the underwater maze at Poseidon's realm, and he will kick aside the rock there, so that you can enter the throne room and get the Trident. Travel back to Mount Olympus where Hercules will break the lock for you. On the mountain, Hercules mistakes Medusa for a snake and gets turned to stone. But if you have the drachma from cleaning the Hades temple, you can get a Wind Bag to take you up to the top of the mountain, where you will battle Zeus.

First get all of the items, then go to the place to the left of main street and then there will be a cutscene where Zeus steals the items. Then go inside the building to the building in main street that has the sign that says Hercules is there. Hercules is on the right. Talk to him. Click the bottom speech bubble thing and he will help you get to Poseidon and Hades. Use the magic mirror from Aphrodite to get to they're places. If you don't have the mirror, then you have to go to Posidon's temple, to the right of Hades', and use the starfish that you will find on Poseidon's statue in the museum, and go inside. Go to the left and talk to Aphrodite. You will have to do a quiz where you have to name different gods. This is why I suggest that you click on all of the plaques under all of the statues.
