There are severnal ways that you can play DVD movies saved as .ISO files on Mac, especially on the on the newest Mac OS Snow Leopard. Use Apple DVD Player or other third party program, such as VLC, Macgo Blu-ray Player etc. Just open the VLC media player and drag the ISO file to the VLC menu. Double click to start playing. An external Blu-ray drive is also needed if you wanna play a Blu-ray Disc.
The DVD ROM will try to read the disc as a DVD or CD and give you a prompt asking what to do. If you open to view files, it will show you the files within the disc. It will not play the disc as a movie or as a game because the computer is not configured to read the files.

As far as i know, the Macgo Blu-ray player allows the Mac player to play Blu-ray ISO on Mac direclty. What's more, the latest VLC can also support the Blu-ray ISO playback, but only the non-commercial ones.
