A veterinarian can ultrasound a mare at 14 days post ovulation and be able to determine if a mare is
pregnant. An experienced veterinarian can palpate a mare that has not returned to heat post breeding (at 25 plus days) and be able to feel the conceptus. A laboratory based blood test can be done for around 100 dollars and there are also home based urine tests that can detect a pregnancy in a mare that is approximately 40-120 days in foal and another if the mare from a 120 days in foal. The urine based tests were developed in New Zealand.
Via ultrasound you can determine pregnancy at 10-12 days post ovulation depending on the quality of the machine. If the mare has been bred and doesn't come back into heat within 20 days of the final breeding date it is
likely she is pregnant. Palpation can be used to determine pregnancy but requires more experience/expertise on the part of the practitioner.
It can be difficult to know if a mare truly is pregnant, however there are some signs you can go by. Starting at about 14 days after the mare was served she should show signs of rejection towards stallions, also if the mare does not begin her estrous cycle 21 days after being served it is a good bet she is pregnant. A trans-rectal palpitation can be done as soon as 16-20 days after the mare was served. Ultrasounds can be used starting at day 16 and can tell you the sex of the foal by day 55 onwards. There are also hormone tests that can be done at varying points in the mares pregnancy to determine if she is still pregnant.
