Isotopes of an element have a different number of neutrons. For example Uranium-235 and Uranium-238 look chemically identical but 238U atom is slightly more massive due to the three extra neutrons.

The mass number is the sum of the number of protons and the number of neutrons. Since every atom, ion, and isotope of the same element will have the same number of protons, the mass numbers of various isotopes are different due to the number of neutrons within each atom.

For example, Lithium has two isotopes: 6Li and 7Li. We also know that both atoms have 3 protons because they are both still lithium. Then all you have to do is do subtraction to figure out that 6Li has 3 neutrons and 7Li has 4 neutrons. This is how you are able to figure out the structures of isotopes and the number of neutrons (defining characteristics of isotopes) through mass numbers.
