The strings on a guitar vibrate when you strum them or pluck them. Depending on how tight the strings are depends on the frequency of the sound (how high or low pitched it is). The circular hole is there to amplify the sound but it is not necessary.

If you get a rubber band and pull it really tight then strum it, it will make a higher pitched sound than if you loosen it and strum it. As you can imagine after time the stings on a guitar loosen that is why you need to re tune it.
