It depends on the exact location of where you plan to start and end, hwever, the distance I saw on Google Maps was 153 miles. This means that it should take about 3 hours to drive from Reno to Yosemite National Park.
The drive from Boise to Yosemite Valley, the main part of Yosemite National Park, is 593 miles, or 11 hours. The nearest entry gate to the park, however, is only about 530 miles, or 10 hours away. If you're not up for the full drive in one day, you could stay in Reno for cheap hotels or in Mammoth Lakes to get up and finish the drive to Yosemite in the morning. (I actually just did this drive!)
