Between 12.5 and 25 minutes, depending on how full the world is (the shorter time applies to a full world). The only practical way of mining runite is world-hopping. However, you will also find a lot of competition from other players, and risk dying because of revenants (if you are free-to-play, the only option is in the Wilderness). You will get about the same amount of money per hour if you mine clay, which only requires level 1 mining!

That varies, depending on the specific ore; for example, clay respawns very fast, while gold takes a long time. It also varies depending on how full the world is; on a nearly-empty world it will take about twice as long as when the world is nearly-full. runescape.salmoneus.net has a table listing the observed and estimated respawn times for different ores.
