If 16 oz. equals 1 pound and the measurements for fresh pumpkin are equal to the canned pumpkin wouldn't that be the same in ounces. So one can is usually 14 or 15 ounces, so you would probably use a little more than one can. I'm only guessing of course, but that is what I would do.
Cups are a measurement for size where as ounces is weight. So the answer varies by brand and product. The best way to figure it out is to look at the back of the can and see the suggested serving sizes which is usually written in cup measurements. If looking to replace a can one and a quarter cups of diced fresh tomatoes and one cup of liquid will do the trick.
