It really depends. These days, many comedians use their time in college to hone and improve upon their "act." For example, Jay Leno attended Emerson College, a school in Boston well-known for performing arts. He performed in local clubs to get more experience. There is no requirement that a comedian graduate from college, but many do, because if they attend a school with a strong program in performing arts, they can be mentored by experienced entertainers. Further, attending college provides a would-be comedian with a back-up plan, which can help him or her to find a job (and some income) in the early years when they are first trying to get hired as comedians.
