It greately depends on the date of issue. Modern issues (last few decades or so) have values between 10 cents and a few dollars, wheras stamps issued before the WW II can have values up to several hundred dollars. The first romanian stamp issued in 1858 sell on ebay for at least 1000 USD.
Without knowing the date on the stamp, a description and condition, it is impossible to value a stamp.
It is called the First Day of Issue. Cancellations on that date have value. Cancellations before that date are really sought after!

It depends on the time frame of the stamp. If there is a special cancellation, you definitely want to leave it on the cover. The older the piece, the more likely the cancellation and date will be of value.
