It can depend on whether or not your area is subject to extreme heat or cold conditions but the usually recommended oil is SAE 10W-30.
The only answer I can find in the manual is when the dashboard "maintenance required" signal appears. I'm thinking it's around 5000 miles, but I don't know for sure, as I just got my 2005 Highlander recently.
It calls for 5W-30 conventional motor oil but in a warm climate you could use 10W-30.
I've drained the trans oil in my 05 Highlander 2.4L engine through the oil pan cap at the bottom of transmission and approx 4.1 quarts came out . I was told there is approx 1 quart left in the torq converter.
