If President Truman autographed the bill himself (i.e. the signature is NOT one of the printed ones at the bottom of the note) you would need to take it to an autograph dealer for authentication.
If you are simply referring to the signatures of the Treasury Secretary and U.S. Treasurer printed on either side of the portrait, neither of those can ever contain the President's name.
Either way, someone would have to examine your bill in person.

Not much, if it is in really good shape, it could be worth about $5 US. There are a few that have some real value. A US Early Reader's edition of Harry Potter and the Sorcerer's Stone is worth about $1,800.
