Of course it will run. But it will not be making full utilization of the multi-core architecture. In that aspect, single-core processors with more processing power are sometimes better suited to run single threaded application than multi-core processors with less processing power.

Pretty sure this motherboard uses the LGA 1366 socket. This slot is used for most of the i3-i7 line of Intel processors, which are all multi-core processors. Yes it will support multi-core processing!
Since the core 2 duo processor has two execution units, it can execute two threads simultaneously. (If you said quad instead of duo, the answer would be four.)
Most computers sold these days have at least two or more cores or microprocessors in them. There are still programs used these days that were written when multi core computers were still a dream. If your computer has multi core machine is running a program that can only use a single core then your program will run as fast as it always has. Where you to get a more recent version of the same program that uses as many cores as your computer has then word processing will be as fast as before but graphic intensive programs such as games make you feel like you have jumped off a pushbike and jumped straight into a sports car - mind the whiplash.
