Yes. One of the provisions of the Adams-Onis Treaty of 1819 is the cession of Florida to the United States by Spain.

The French was the previous owner of British Cession. The United States purchased the western half of the Mississippi basin from France.
Louisiana, Alaska and the Gadsden Purchase were the only true purchased areas. Florida and the Mexican Cession/Texas could also be counted as purchased, though the US gave the money for Mexico and Spain's losses. The Mexican Cession and Texas were truly won through the Mexican-American War and Florida was truly won by Andrew Jackson taking over the government, then Spain basically handing it over in fear of a war.

Spain ceded Florida to the US in 1819 when James Monroe was President. I am not aware that James Madison had anything to do with the transfer.
