IF, and only IF you are able to keep the hot Poker on it for sufficient time to actually give your self a 3'rd or maybe even 4'th degree skin burn at the place where the mole is, then yes. It can be done. I did not say it was without pain, and I did not say it is safe.
You are at high risk of getting an infection afterwards. Burn wounds are quite different from other types of wounds. They heal very very slowly and will leave a great big scar.
A better way would probably be to locally freeze it down. This should give much of the same effect but does not scar as much. It is painfully but not by a long shot as painfully as the hot poker.
I am sure a doctor can help you to get the mole removed professionally without it costing an awfully lot.
If you mean burn like on fire, then yes, I think it can (but it might explode).

You might think a flare would not burn in a vacuum, however, they will. The oxygen for the flare to burn is already chemically in the flare. And flare guns HAVE gone into space- as emergency equipment.
