If you put a salary requirement in a cover letter that might be too high is it OK to call or email and let them know that a lower range is acceptable?

You definitely run the risk of sounding desperate.
Ideally they'd see your resume and contact you to ask if you'll accept a lower salary. However, if you don't hear from them you probably have nothing to lose.
