In "The Original Series" Mr. Spock was already onboard as the Science Officer; both the Pilot episode, and the released series. In the latest movie, Kirk met Spock when he 'cheated' at the "Kobayashi Maru" test, or at the trial quickly following it.

Because the crew of the Enterprise is military, often they are all referred to by their last names, but Vulcans refer to them by rank or title when needed, and by first name in friendship situations. Spock called Captain Kirk Jim, and even in the new movies, Spock said his girlfriend's first name (which Kirk didn't know).
