Although both of them are insane in some form or another, the Joker's insanity isn't usually a source of humour but rather the thing that makes him a good evil counterpart to Batman. The closest DC equivalent to Deadpool would probably be Ambush Bug because both of their humour is very meta, stemming from the fact that they are aware they are in comic books, including being able to see the speech bubbles, mechanics of the comic book universe, and so on. This extends to games and (presumably) to screen appearances as wll.

Hmmm... well that all depends on how seriously Deadpool takes the Joker. If Deadpool started laughing at the Joker's appearance, he would probably be either killed, or severely injured. If he took the Joker seriously, he would have a pretty fair chance of winning if he keeps his distance. The Joker has a plethora of deadly short ranged weapons. As far as I know, the only long ranged weapons he owns that could possibly hit Deadpool would be his playing cards, his harpoon gun, or his gun. However, since Deadpool has superhuman reflexes and speed... I'd say only the playing cards could possibly hit him, but damage would be minimized due to Deadpool's amazing healing ability. Lol sorry if I sound like a total geek here :P
