Three of the Clemens children died before marrying - Lampton, Susan (Susy) and Jean. Clara did marry and had one daughter, Nina, who is dead with no children. There are no living descendants of Mark Twain.

Update: There is now DNA evidence that Mark Twain does have a great granddaughter, child of his granddaughter, Nina Gabrilowitsch. That book has now been published and is on Amazon.

No, Mark Twain died April 21, 1910 from heart failure one day after the passing of Halley's comet. He is quoted saying,

" I came in with Halley's Comet in 1835. It is coming again next year, and I expect to go out with it. It will be the greatest disappointment of my life if I don't go out with Halley's Comet. The Almighty has said, no doubt: 'Now here are these two unaccountable freaks; they came in together, they must go out together.'"

Clara Langhorne Clemens Samossoud (June 8, 1874 - November 19, 1962), was the daughter of Samuel Clemens, who wrote as Mark Twain. She was his last surviving daughter.

Let's see if I can improve on that answer. While it's true she was his last surviving child, she married Ossip Garbilowitsch, a Russian musician and director of the Detroit Symphony Orchestra. They had one daughter, Nina, born in 1910, and some people believe that it has been proven by DNA that Nina had a daughter. So that child would be the closest direct descendant of Mark Twain.
