Unfortunately there is not enough information to give a thorough answer. If this man and his co-worker are normally up at this hour, the call was only ten minutes, and this is not common. Then no, the man is certainly not cheating.
If he's not normally up at that hour, the calls start running longer, he only talks to the co-worker while his wife is at work, and it goes on several days out of the week, then there might be some cause for concern, but it doesn't automatically mean he is cheating.
