If the test is ordered through the courts, you can actually go to jail for refusing to establish paternity. So, no.
If you are a legal guardian of the child then you can take the child and go get a paternity test yourself.

Rather, electrophoresis is merely one of many analytical techniques which may be applied to a specific question, being posed by a an investigator. The value of electrophoresis is nil outside the data quality objectives (DQOs) presented by the investigating team, and the QA/QC program of the analyzing laboratory. Without QA/QC, in the lab, and without appropriate DQOs, electrophoresis is no more capable of implicating a suspect than a ouija board. Laboratories cannot perform in the real world as seen on TV's CSI, and real CSIs would go to jail if they pulled some of the stunts seen on the TV series.

Children receive half of their genetic material from each parent. There are specific sites on DNA, known as restriction sites, that are recognized by restriction enzymes. These are used to determine paternity. Samples of DNA from the mother, father and child are taken. They are all digested ('cut') by the same restriction enzymes. These DNA fragments are then separated by gel electrophoresis (which separates fragments based on size). The bands of the child are compared to the mother and father's. If the band is not the same as the mother's, it must have come from the father. If these do not match up, then the sample was not taken from the biological father.

Paternity tests can give a family answers when paternity is in doubt. Once the test is conducted, all paternity questions will be resolved and the family can heal.
You can only "amend" a paternity affidavit if you have a paternity test done to prove or disprove paternity... In many cases this is something that the state will do (ie of child support orders)
