Depends on what you're talking about. If you start a tweet with @username, only that person you mention will see it. Now, if you place @username anywhere else in the tweet, everyone who follows you will see that tweet in their timeline.

This isn't possible as the idea of your timeline is that when you post a message, those that follow you will see it. You can only send tweets to individual members by 'mentioning' them (@username). If you really wanted to then you could send the same tweet to everyone you follow but this would take you a long time and it's much easier to post it to your timeline. If you want someone you're following to see your tweet and they aren't following you then you should mention them in your tweet. You could even ask them nicely to follow you back and then you wouldn't have to do so in future.
