The tattoo is a wound with a scab. This blister you speak of is a layer of the scab and should not be picked off or brushed off or messed with in general. It's all part of the healing process. If it's bothering you, itching excessively, or flaking off too soon, apply some dye-free and fragrance-free lotion to it.
