If it's a modern US bill, the back and front should be oriented the same direction. That's different from US Coins whose sides are oriented 180º apart.
If your bill's reverse is upside down (i.e. the bottom of Independence Hall is along the same edge as the top of Franklin's portrait) you could have a moderately valuable printing error. In any case you should have the bill evaluated in person by a dealer or appraiser who works with error currency.
