However both had a secondary purpose as weapons effects tests to determine actual type and amount of damage produced by the new bombs compared to use of massive firebombing attacks with conventional bombs. This was why they selected target cities that had not been significantly bombed already and then the cities on this target listed were then made "prohibited firebombing targets", to make estimation of damage due to the atomic bombs simpler.
A tertiary purpose was to verify yields: the MK-I yield was not known as it had never been tested before use and the MK-III yield although known from the Trinity Test needed to be confirmed if it was reproducible from bomb to bomb.

Testing of this weapon was considered unnecessary, and fissionable material was very scarce at any rate. The Nagasaki weapon, an implosion type weapon called "Fat Man", was so complex that testing was imperative. It was tested at Trinity site in July 1945,& the bombs were dropped on Japan a few weeks later. So, yes, one type of bomb was tested, one time, before they were used on Japan.
