Jamestown was founded first, and was a financial venture (they hoped to make money) Plymouth was settled for religious reasons. First settlers at Jamestown were all men, some of the first settlers at Plymouth were women.
Both Jamestown and Plymouth were English colonies, and were formed in the early 1600's. Both colonies were helped by Native Americans. The pilgrims believed they were not bound by the rules of the Virginia Company. however, they needed rules of some sort so 41 adult men created the mayflower compact this was to be used for just and equal laws. office holders would be elected by the colony's adult males.
Plymouth holds a special role in American history. Rather than being entrepreneurs like many of the settlers of Jamestown, the citizens of Plymouth were fleeing religious persecution and searching for a place to worship their god as they saw it.
