Engine "knock" is preignition or detonation of the air-fuel mixture before the piston is in power stroke position. It is usually caused by a lean fuel mix. So to check for preignition, check your plugs for signs of lean running.

What does a knock sencor do?

A knock sensor listens for the 'knock' (or pinking), that shows that the fuel is exploding, rather than burning. Via the engine management computer, it retards the timing of the spark, until the knocking stops. 'Knocking' can damage valves and piston crowns, if allowed to continue. A knock sensor allows different grades of fuel to be used, without damaging the engine.

Have a 97 Olds 88 replaced MAP sensor and new gaskets for coolant leak everything was great. Now about 250 miles later the engine has a loud ticking knocking noise. Would this be a knock sensor?

The knock sensor is exactly what it says. It senses any knock or ticking in the engine and retards the timing. The knock sensor is not the problem, the knocking is. Go to a repair shop and have it diagnosed.
