The first four Caliphs are all informed by Prophet Muhammad (PBUH) that they are promised by God to among ten prophet Muhammad companions who to be accepted the God Paradise in their 2nd life. Also, the four have close relations to prophet Muhammad (PBUH). Prophet Muhammad (PBUH) married the daughter of each of the first two Caliphs. The third Caliph married the daughter of prophet Muhammad and when this daughter died he married the other daughter. The fourth prophet married Fatima (may God be pleased with her) the youngest daughter of the prophet.

There was an attempt at poisoning the prophet (pbuh). It was a Jewish woman from the tribe of Qurayza, after the men had been killed. She attempted to poison the prophet, but she was not successful, even though the prophet had a taste. Once he realized it was poison, he called for her... And asked why she did it. She gave him the reply, " You killed my husband, and my father, if you are a true prophet you would surely have realised it was poison, if you are a fraud, i would have favoured the people by getting rid of you" He then forgave her, and let her free. He died four years later, and as you know.. no poison takes 4 years to kill a man. So he was not poisoned.
This is kind of like saying who was the successor to moses. Muhammad founded Islam and taught the people about it. But he wasn't in an official position that could be replaced, he was a prophet.
The beloved Prophet Muhammad (May peace be upon him) had four daughters. All of them died natural death. However, the eldest Hazrat Zainab (RAU) was seriously wounded when she tried to migrate from Makka to Madina. Anyhow, she didn't die at that time. If somebody reports that any of the four daughters of The beloved Prophet Muhammad (May peace be upon him) was killed by any means or poisoned, he tells a lie. No wretched unfortunate Muslim can think of such a hellish act. Having highest regards and love for the children and grandchildren of The beloved Prophet Muhammad (May peace be upon him) is the basis of every Muslim's faith and Eeman.
Some believe Prophet Muhammad (peace upom him) had four daughters; namely Umm Kulthum, Ruqayah, Zainab, and Fatimah bint Muhammad. But a large percentage of Muslims also believe he only had one, namely Fatima. Looking from a mathematical and physiological perspective, it is unlikely he fathered more daughters after Fatima.
