malware scanner--malwarebytes, superantispyware (disable real time scanning to not conflict with your antivirus and run manual scans occasionally)
Keep your computer up-to-date. This includes your antivirus and really everything on your computer. Many infected computers are infected because of security issues that could be have been fixed.
Hijackthis--download this and post log to a forum if you suspect invasion. This is probably the most surefire way to remove any virus or malware. Basically it logs the stuff going on, on your computer, you post a log, and experts analyze that log and tell you what to do. Most of them will tell you to download malwarebytes/superantispyware and to scan and repost your log.
Firefox or Opera browser--disable IE by restricting it with your firewall. You can unrestrict if you need to use it for something but otherwise I would use Firefox (personal preference over Opera...sorry Opera people I know it's great). Noscript for the paranoid.
Safe browsing--prevention is worth a pound of cure. If you are a bit torrent, youtubing, online gambling fool...well then you will probaby get infected eventually.
Hosts files--Google mvp hosts file
Blockers--Spybot search and destroy is useful for immunizing your computer. As is spyware blaster. Disable teatimer for spybot so as not to conflict with antivirus.
Online scanners--There is a host of online scanners that you can use for one time scanning of your computer. Among the top are trendmicrohousecall, Nod32, Windows One Care.
Strong passwords--I use very strong passwords that are ridiculously long and write them down. They are different for all sites. I then have a few digits/symbols that are common to all my passwords that I don't write down in case the sheet were stolen. In other words if my password were 33d@f87*$#832jk782jk92o&ilm9%b I might right down @f87*$#832jk782jk92o&ilm and make 33d9%b common for all my passwords (the first and last three digits). It basically renders the sheet useless. I also don't right down what the password is to on the sheet. For bank I might write a capital b.
