Though Gettysburg is often touted as the turning point of the war, the capture of Vicksburg which effectively closed the Mississippi to Southern boat traffic is actually the most decisive strategic achievement. It completed phase two of the Anaconda Plan.

This is open for debate, but a good place to look would be the Death of Stonewall Jackson followed by the battles of Vicksburg and Gettysburg. This marked the end of a seemingly invincible Confederate army and the last Confederate push into the North. Up to this point, the Union was on the backfoot. Afterwards, the Union troops regained the momentum and pushed the war into Confederate territory.
Vicksburg - it liberated the Mississippi, denying the enemy all use of this important military highway, isolating all Confederate units to the west of the river, and enabling Grant to go to the aid of the Army of the Cumberland.
Gettysburg, which ended on the same day, was the more dramatic moment, but it held less significance, being mostly a psychological body-blow to the Confederacy, and to Lee himself, whose health started to decline from this point.
