I'll hazard a guess you're asking about the brake warning light on the dash, not the brake lights at the rear of the vehicle. The warning light will come on if brake fluid falls below a certain level, if a fault is detected in the master cylinder, if there's unequal pressure in the brake lines (usually due to a leaky caliper) or other issue. It might also simply mean your emergency brake is applied or the switch is bad.
Whatever the cause, don't ignore it. If you're not mechanically inclined, take the car to a brake shop, your favorite mechanic or a Toyota dealer. Brake failure is not an option.

If a brake light stays on it's usually just a sticky or bad switch. The brake light switch is under the dash and is activated by the brake pedal. It could have some grunge on it preventing the switch from closing. Try cleaning the plunger. If that doesn't work (and it probably won't) replacement is very easy. Cheers
you may have your brake light socket and your back up light socket in the wrong holes. Or you have a short but that would probably leave you with a blown fuse and no lights.

The lamp in the top brake light of a Toyota Highlander can be replaced by the locating the light housing from the trunk. Find the light cover and remove it to access the lights. Replace the bulbs.

More than likely your brake fluid is low. Go buy a small can of DOT3 or DOT4 brake fluid and top it off. The reservoir is located near the firewall in the engine bay, on the driver's side, and has a rubber cap that snaps on/off rather than a screw cap.
