There are three verses that talk about the location: Mark 14:15 says, "And he will shew you a large upper room furnished and prepared: there make ready for us." Luke 22:12 says, "And he shall shew you a large upper room furnished: there make ready." Acts 1:13 says, "And when they were come in, they went up into an upper room, where abode both Peter, and James, and John, and Andrew, Philip, and Thomas, Bartholomew, and Matthew, James the son of Alphaeus, and Simon Zelotes, and Judas the brother of James." That is all the Bible says about the location.

In the upper room of the temple* in Jerusalem.

* The upper room, or Cenacle, was not part of the temple but was part of a private home that was borrowed by Our Lord for the Passover meal. It took place on a Thursday evening during the Jewish Passover, the day before Our Lord died on the cross, about the year 30-33 A.D.
