I am reading a book about him by Chris Mathews and when JFK got sick they didn't know what he had. It took until he was an adult to figure out it was Addison's disease . This is a form of leukemia . He also had a bad back and off and on through his life he was never well. They thought he would die 3 times. As he began his organizing for his 1960 run for president he is quoted as saying that if he was 80-85% well that was all he could expect. Considering his health problems it is possible he could have died in his second term.
