First make sure you have all the stars from all the courses (you can do this by pausing the game in the castle and using the arrows to see what stars you have in each course), including the 100-coin stars. Also make sure you have all the secret stars and their red coins (if any). There is a red coin challenge in Bowser's last level, so that must be the one your missing.
When you get the 150th Star in Super Mario 64 DS, the cannon in the castle grounds will be unlocked. This will allow you to get on top of the roof of the castle, where there are 3 1-Up Mushrooms, a ? block and a rabbit (for Luigi).

Firstly, there aren't even 150 Stars in Super Mario 64. The DS remake has 150 stars.Secondly, you only need 70 of them (80 on the DS) to actually beat the game. Once you get that number of stars, you will be able to challenge the final boss, and beat the game.
