If a guy doesn't respect you as a virgin, or thinks you're a prude, don't even bother. Being a virgin is a very good thing. You don't have to be an idiot to believe it's something worth keeping until the right time. Don't lose it to somebody you don't 100% love. Make sure this isn't one of those mistakes.
Tell him you are a virgin if it comes to that point in the relationship. Be honest, and watch his reaction CLOSELY. If he thinks it is a good thing, and is sincere, then you have a good man. If he doesn't like it, or tries to pressure you to do more and won't listen to you about wanting to wait, then he is not the guy for you.

If you're engaging in any kind of intercourse, yes, if they've had a lot of experience they'll be able to tell because you'd be tight and most likely experience pain. But, if you're just hanging around, kissing, making out, dating etc then odds are, no. and there is a thin layer of membrane in the entrance of the vagina called the hymen, if it hasn't already been broken than yes, he will know because when you have sex it breaks it, this is also known as "popping the cherry"
