To listen to this ' a joke is a very serious thing' verbally, it may sound wrong or because a joke is a contradiction to seriousness. But as I tried to really find out what it means, what I concluded with was.. life is a longrun and two things he tackles: seriousness and joke (humour).. And these two things doesn't come together but rather it substitutes.. thus this could mean the equal importance of the two.. to make life meaningful, beautiful, enjoyable and go on relentlessly.

To reader formal is serious and is not very happy, like a letter from the government to some kind of bank. An informal is funny and not serious like someone giving a birthday card which has a funny joke in.
