This seems like a very generalized question so I'll answer it in general terms too.

GUI = Graphic User Interface

What is meant by the term Interface in this acronym is the interaction between the peripheral devices (keyboard, mouse, etc.) and the computer processor.

It interfaces or allows the user to communicate with the remainder of the hardware through the Operating System.

Graphic just means it isn't done using command line.
