Holy Water is mostly known in the Catholic Church. I am unaware of Protestant sects which use Holy Water.
Holy Water is a "Sacramental." Sacramentals are signs, they point to one or more of the seven Sacraments. Holy Water therefore is a Sacramental of Baptism. When people use it and bless themselves with it, they are reminded of their own Baptism whereby God made a covenant with them.
Holy Water is used in cases of possession because it is a blessed object. It can often times be used in conjunction with blessed salt. When an object is blessed, that simply means that the object is set apart for use in sacred rituals, and the power of Christ is connected with the object. Holy Water is spiritually cleansing and therefore makes the demon uncomfortable when used in cases of possession. I must admit I do not know much about this subject, but I do know at least what I have written above.
It is important to note that in cases of possession or in cases whereby a house is "haunted" no person should attempt to perform an exorcism ritual on their own. I also advise people to NOT consult "Spirit Guides" or "Wiccans" or any other new age type person but a Catholic Priest Exorcist. This is not to say that "Wiccans" or "Spirit Guides" or New Agers do not have good intentions, but it is to assert that casting out demons is only done through the power of Christ. Attempting to cast out demons under the authority of any other name or power then that of Christ will have disastrous consequences. Failing to follow this advice can possibly have the consequence of making things far worse. It is also important to note that a Catholic Priest Exorcist cannot charge for his services. The ministry of exorcism can only be done out of the motive of Charity.
It would depend on your beliefs, but many think it can help to expel any demon because of its holiness. However, some who believe in demon possession have disbelief in holy water as something helpful, and believe strong faith is all that's needed; no objects or things deemed holy by man can do anything if their is no faith.
Holy water does nothing in this case, because possession by demons is no more than a superstition. There are those who are willing to take advantage of the superstitious, either to benefit the Church or for financial gain, but holy water and exorcisms do nothing real.
To help a demon possessed person, there are several steps that can be taken and this all really depends on your religious beliefs. For a Christian it's safe to pray and ...pray...and pray and throw holy water on them. For an other, burn sage and walk around the house expelling the demon in your own way. Then place a circle of salt around your place of dwelling.Answer:Find some one with the Baptism of the Holy Spirit to pray for them, and in the Name of Jesus Christ demon must leave. If you don't have this gift, do not do this yourself.

Warning! Spoiler for the last season! No. Although there were episodes, where demons possessed Sam, he is not a demon. The possessed people are only vessels. And although Lucifer possessed him at the end of the last season, he is an Angel. An Angel in the vessel of a demon seems a little unnatural.
