Think about it for a minute. Lions are (a) very territorial and protective of their dens and (b) capable of killing a human with one paw-swipe. So, "walking into a lion's den" would be taking some action that puts you at great risk of destruction.

Today the phrase is more associated with an action that puts you at risk of some social or business destruction. For example, a person who sent an e-mail flame to his bosses might be called in to a meeting with all of them to find out what happened.
