The event that conflicted them is that the Jamestown settlers took the Powhatan leader and killed him. So this made them gom in wars.
The story we know is not true. Actually the colonist landed in an area of 14,000 Native Americans and in the worse land in the area. Powhatan pretty much left them alone. I think he figured that they would die from the bad water and disease . He was fairly right. Within 6 months there were only 34 men left alive of the 104 who came. It wasn't until after his death that the brother of Powhatan attacked the colony. The story about Smith is also not true. He did NOT save Jamestown. He was only there a very few months and he lied about his contribution to the settlement in a book he wrote several years later. He also made up the story about Pocahontas and she died young so couldn't refute his story. The man who did save Jamestown was the husband of Pocahontas and gave tobacco seeds to the colony. That was John Rolfe.
This is one thing that the Disney Film gets 100% right. When asked why the Indians are attacking one of the characters says "Because we invaded their land and cut down their trees and dug up their earth?" Even in real history that is 50% of the reason for the conflicts. The English simply showing up and building then not leaving.
What lead to the other conflicts could be called trade disputes, then eventually land disputes.
The English colonist who came at first were looking for a quick score. They wanted to dig up some gold and go back to England. So they did not plant crops and they ended up relying on Powhatan for corn and other food stuffs in trade. Sometimes the trade was not always peaceful, one side or the other felt cheated or slighted or actually was cheated or slighted.
Latter on once Tobacco cultivation became the main business of the colony wars were about land. The Indians would sell various parcels to the English. Eventually some English would simply squat on the Indians lands. This would spark conflicts off. The final "Anglo Powhatan" war ended in 1646 with the English taking a defined wedge of land away from the Powhatan. Borders were defined and a system of passports was established between them.

The Powhatan basically left Jamestown alone for the first few years because the knew that the fort was built on swampy land with mosquitos that carried diseases and they had bad water. The colony actually began in an empire of 15,000 Native Americans and were also out numbered. Because of the conditions the Powhatan knew that the Jamestown men would die of disease and starvation . They were right. One hundred and four men landed and in six months only 34 were alive and the Powhatan didn't have to do anything. Much later as the colony did take hold because of tobacco there were problems and a massacre by the Powhatan , but that took years to develop.
The Jamestown settlers forced a crown on Powhatan and made him swear allegiance to the British King, starting strained relations.
They lost their land.
Some sources, such as http://etext.lib.virginia.edu/etcbin/jamestown-browse?id=J1023 suggest that when the Europeans first settled in Jamestown, the relationship they had with the Powhatan Indians was very peaceful. The Indians helped them to settle by offering food such as Indian cakes and corn. Other sites, such as http:/en.wikipedia.org/wiki/Powhatan#History suggest that the violence started right away. Either way, there was violent interactions between the European settlers and the Powhatan Indians that led to mass murdering of both sides. Personally I would be more likely to believe the first link because it's a letter from Edward Maria Wingfield who was one of the very first to settle in Jamestown.
The Powhatan traded fur and food with the settlers.
Pocahantas helped make peace between The Jamestown colonists and the Powhatan because she had married the English colonist John Rofle in the year 1614. John Rolfe wasn't a colonist!! :)

Chief Powhatan was being a very gracious person to the colonists but the colonists never showed that they were thankful or if they appreciated their help...and as Jamestown receives a new leader (John Smith) things got better.(This answer explains nothing.)
The main reason that conflict increased between the Virginia Colonists and the Powhatan Indians was because the Powhatan were being pushed off their lands to accommodate the Colonists. Then Captain John Smith was captured by the Powhatan Indians and Pocahontas saved his life.
