It depended on which businesses you were asking about. The top 1-2% of the upper class was mostly untouched by the Depression. Giant corporations and those that sold luxury items continued to make a profit, as did radio networks, and even movies. But as for everyone else, it was a harsh period of time that resulted in massive closures of stores, banks, and factories nation-wide. Smaller businesses in particular were severely affected; because millions of people were unemployed, they were unable to afford to shop, and that loss of customers had a ripple effect, causing a number of local store owners to either close their doors or in some cases, declare bankruptcy. When the economy gradually began to turn around, some businesses did re-open, but many were never able to do so.
