If El mustachio escapes through the tunnel just continue to do the game and you will eventually catch him in the end. Unlike me because when I got him with my lasso he escaped before I got to diamond Plains.

You get the clock working then the train should pull up. Get on the train and shoot El Mustachio and his gang to scare them off. Then you go to there secret hideout, use your invisibility potion to go in the shed. then shoot things to distract them and shoot everyone but El Mustachio. Then El Mustachio will ride out on his horse. Chase him until he reaches a dead end. Lasso him and bring him to the Dimond Plains ranch. Hope this helps!
