That usually depends on how long you were on the depo. The longer you were on it (like over a year vs 1 or 2 months the less likely you would be) But if you had sex around 5 days late for injection you might want to have a pregnancy test before your next shot.
No, actually my nurse has told me that it is better to get on the depo when you are on your period. It causes it to stop, and it is effective quicker.

You can get a Depo Provera shot whether or not you're having vaginal bleeding. There is no medical reason to prefer one time over another.
You can get Depo Provera at any time in your cycle. Getting the first Depo Provera during your period is easier because it becomes effective faster (within 24 hours) when you take it in the first seven days of your period. If you get it at any other time in your cycle, you'll get a pregnancy test at the time of the injection. You should repeat the pregnancy test in two weeks. You also need to use a back up method of birth control for seven days.
