DNA ligase plays a very important role in rDNA technology.It seals the single strand nicks in the double stranded DNA.It forms a phosphodiester bond between the two nucleotide.Based on the type of energy they use during ligation DNA ligase are of two types:

1.E.coli DNA ligase(which uses NAD as a source of energy) and

2.T4 DNA ligase(which uses ATP as a source if energy)

The ligase enzyme glues the pieces of DNA together. It's normal function is repairing double strand breaks in DNA molecules.

When you digest a plasmid, for instance you cut it at certain base pairs. Ligase is the enzyme that can " paste these cut ends together into whole plasmids with differing genetic complements. It is also useful for other forms of DNA.

DNA ligase is an enzyme which seals gaps in the sugar - phosphate backbone of DNA molecules.

This is necessary in the replication of DNA as the lagging strand is synthesised in short fragments by DNA polymerase. The gaps between the fragments are then sealed by DNA ligase.

DNA ligase is also very important in genetic engineering. When a plasmid (circular strand of DNA) is cut open and an extra section of DNA inserted, it is DNA ligase which seals the two DNA molecules together.
