Sickle cell anemia is caused by a single changed amino acid in the beta subunit (there are two of them) of the hemoglobin protein from a glutamic acid to a valine. This change causes an exposed hydrophobic region on the subunit that ends up locking onto the hydrophobic region of a subunit of another hemoglobin and so on. The continuous locking causes the molecules to crystallize into a fiber and the capacity to carry oxygen is thus greatly reduced. The hemoglobin fibers deform the red blood cell into a sickle shape.

Normally red blood cells are round and doughnut shaped (without the actual hole) and so, pass through comfortably through the blood vessels. The sickle cells are stiff and sticky, not at all able to continuously pass comfortably through the blood vessels. They tend to clog up in the vessels, blocking flow of blood to the limbs and organs. This can cause pain, serious infections, and damage to vital organs.

Sickle cell anemia is just one type of anemia. Anemia occurs when your blood has a lower than normal amount of red blood cells or when your red blood cells don't contain enough hemoglobin. In the case of sickle cell anemia, the hemoglobin's capacity to carry oxygen is drastically reduced, thus creating the same conditions of anemia.

It is when a mutation occurs in your DNA sequence, causing a mess up. Because of this mess up, it changes your normal red blood cells into sickle cells. Normal red blood cells are able to squish and manuever easily through our organs to transfer the oxygen, but sickle cells are pointy and can easily get stuck. When all these cells get stuck, it causes blood clots.
