Sickle cell anemia is caused by a single changed amino acid in the beta subunit (there are two of them) of the hemoglobin protein from a glutamic acid to a valine. This change causes an exposed hydrophobic region on the subunit that ends up locking onto the hydrophobic region of a subunit of another hemoglobin and so on. The continuous locking causes the molecules to crystallize into a fiber and the capacity to carry oxygen is thus greatly reduced. The hemoglobin fibers deform the red blood cell into a sickle shape.
