In .NET all the lanuages use a runtime compiler known as CLR (Common Language Runtime). All the languages wishing to target CLR need to comply with CTS or Common Type System. CLR compiles the code written in .NET based languages to IL code or Intermediary Language or MSIL. MSIL is a set of instructions that can be compiled into native mahcine level code as a second and final step of compilation.

C#,C++, and Visual Basic are the languages typically used but there are other .NET compilers for python, ruby, etc. It is a framework and it's suports multiple languages
VB6 code is directly compiled by VB6 compiler into native code. However C# code is targeted for CLR and goes through two level of compilattion. It is first compiled into MSIL and then JIT compiled to native code. This ensures interoperability among .NET based lanuages and hence language independence for them.
