If you register a copyright, that means you've created something and want to protect your rights to it. You can then license others to use that work. So copyright registration is saying "this is mine," and copyright licensing is saying "but you can use it."

Licensing a work means the rightsholder allows a certain specific use, often for a limited time. A single work can be licensed many ways to many people for many purposes, and the original rightsholder retains all the rights granted by the law.

Assignment is the (often) permanent transfer of copyright to another entity. The original rightsholder surrenders their rights to the new copyright holder.

Not really. Registration used to be the way to formally protect your work through copyright. Registration is not required for protection anymore: a work of sufficent originality is automatically protected by copyright as soon as it is fixed in a tangible medium. Some countries and private companies offer registration separately.
