The stroma of a plant is the region outside of the thylakoid space. This serves two functions in the both the light-dependent and light-independent (Calvin cycle) reactions. In the light-dependent reactions, the H+ ions that build up within the thylakoid begin to form a concentration gradient between the thylakoid and the stroma. As a result, the H+ ions need to diffuse into the stroma. In order to do this, the ions must travel through an enzyme known as ATP synthase. Once it does, the movement of H+ ions through ATP synthase into the stroma provides energy for ADP to become ATP. The ATP formed is an energy source to power the Calvin cycle. Now, the light-independent (Calvin cycle) reactions occur in the stroma.

The purpose of the Stroma (Colorless liquid between granum) is, at the most basic level, to commence the the Calvin Cycle. This cycle is rather complex, so if youre taking an AP Biology course or a college Biology course this should be studied in stronger detail, but here is an easy to understand explantion.
1. Before the stroma has a purpose, the first steps of photosynthesis must take place in the Thylakoid (steps 1-5)

3. Light energy is used to split h2o. Thus, releasing o2 (which will be released out of the cell), H+ which will go into the lumen, and a free electron which will be explored later.
4. The e- is pushed through an electronn transport chain,powered by light, through proteins. These e- produce more h+ into the lumen.
5. These H+ ions will then pass through a protein called ATP-Synthase creating ATP and NADPH. This protein releases these molecules into the stroma.
6. Now, the Calvin Cycle begins in the stroma
7. The NADPH and ATP from the thylakoid provide energy in the Calvin Cycle. Co2 is ultimately made into G3P ,through the use of enzymes, which can easily be made into monosaccarides or polysaccarides but is also reused in the Calvin cycle. This is also known as Carbon fixation.
8. Some bonus information, if there is no CO2, then O2 can take its place. This, with the help of RuBisCo, the enzyme also used to make G3P, will create 3GP which plants CAN NOT USE! This phosphoglyceric acid is not good! Remember, G3P is reused to restart the Calvin Cycle!
