That very much depends on what you think of as "one ride". There are people who've spent years touring country after country on bicycles, covering thousands of miles. There's a type of riding called randonneuring, and those guys will happily do 300+ miles in one go, only stopping for food and toilets, and not sleeping on the whole trip.
There are quite a number. The most famous is probably the Tour de France (about 3 weeks, over 2000 miles (generally 100 miles+ per day) through France and sometimes surrounding countries.) There are a number of other professional races like that (Tour d'Italia through Italy, Tour of Spain, etc.) One long continuous race in the US is the Furnace Creek 508, through Death Valley in California. There is also the RAAM (Race Across America), a continuous race (no stages, just go as quickly as possible) from California to Atlantic City, NJ.
Most famous is probably the Tour de France, or maybe Giro di Italia. Most difficult might be Paris-Roubaix. Although much shorter, the riding conditions are much more difficult.
But then there are the endurance races; Paris-Brest-Paris(PBP), or London-Edinburgh-London(LEL). These are every bit as extreme in their own way, if not more.
